Nitrogen, energy and vitamin nutrition of Rhizobium japonicum.

چکیده
The energy, nitrogen and vitamin nutrition of 36 strains of Rhizobium japonicum were investigated. Most of the strains utilized gluconate, glycerol, arabinose, glucose, mannose, mannitol, sorbitol, galactose and maltose. Pyruvate, succinate, ribose, ribitol, xylose, fructose and lactose were of limited use whereas acetate, propionate, lactate, malate, citrate and sucrose were uniformly inadequate as carbon or energy sources. All strains grew well with vitamin-free casein hydrolysate, and 6 could utilize ammonia as sole nitrogen source. Glutamate was utilized by 33 strains, histidine by 26, aspartate by 20 and proline by 26 strains. Other amino acids were not utilized by most strains. The organisms grew independently of any vitamins except biotin. Three of the strains were stimulated and growth of 4 was inhibited by biotin. Received 26 February 1968

Conservation of symbiotic nitrogen fixation gene sequences in Rhizobium japonicum and Bradyrhizobium japonicum.
Southern hybridization with nif (nitrogen fixation) and nod (nodulation) DNA probes from Rhizobium meliloti against intact plasmid DNA of Rhizobium japonicum and Bradyrhizobium japonicum strains indicated that both nif and nod sequences are on plasmid DNA in most R. japonicum strains. An exception is found with R. japonicum strain USDA194 and all B. japonicum strains where nif and nod sequences...

Soybean (Glycine max [L.] Merr. cv Hobbit) plants were grown in a growth chamber for 56 days in a phosphorus- and nitrogen-deficient soil and were colonized by the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus mosseae (Nicol. & Gerd) Gerd. and Trappe and Rhizobium japonicum strain USDA 136, or by either organism alone, or by neither. Non-VAM plants received supplemental phosphorus and no...
متن کاملOccurrence of choline and glycine betaine uptake and metabolism in the family rhizobiaceae and their roles in osmoprotection
The role of glycine betaine and choline in osmoprotection of various Rhizobium, Sinorhizobium, Mesorhizobium, Agrobacterium, and Bradyrhizobium reference strains which display a large variation in salt tolerance was investigated. When externally provided, both compounds enhanced the growth of Rhizobium tropici, Sinorhizobium meliloti, Sinorhizobium fredii, Rhizobium galegae, Agrobacterium tumef...
متن کاملDifferentiation of Rhizobium japonicum, I. enzymatic comparison of nitrogenase repressed and derepressed free living cells and of bacteroids.
Derepressed free living cells of Rhizobium japonicum strain 61-A-101 with leucine as single nitrogen source develop a maximum nitrogenase activity of 180 nmol C2H4.mg protein -1.H-1 in liquid culture under 2% 2% O2 in the gas phase. Only 10% of this activity is found with no oxygen in the gas phase during a 90 min incubation period. The maximum activity under 2% oxygen in the gas phase is unaff...
متن کاملRelation between Glutamine Synthetase and Nitrogenase Activities in the Symbiotic Association between Rhizobium japonicum and Glycine max.
The activity and extent of adenylylation of glutamine synthetase was examined in both free-living and bacteroid forms of Rhizobium japonicum in the presence of excess ammonia. Ammonia caused an apparent repression of glutamine synthetase in free-living R. japonicum and adenylylation of the enzyme was also increased. In contrast, neither the activity nor the extent of adenylylation of the bacter...
